Throughout the world, coastal resource managers are encouraging the restoration of previously modified coastal habitats back into wetlands and managed ponds for their ecosystem value. Because many coastal wetlands are adjacent to urban centers and waters used for human recreation, it is important to understand how wildlife can affect water quality. We measured fecal indicator bacteria (FIB) concentrations, presence/absence of Salmonella, bird abundance, and physico-chemical parameters in two coastal, managed ponds and adjacent sloughs for 4 weeks during the summer and winter in 2006. We characterized the microbial water quality in these waters relative to state water-quality standards and examined the relationship between FIB, bird abundance, and physico-chemical parameters. A box model approach was utilized to determine the net source or sink of FIB in the ponds during the study periods. FIB concentrations often exceeded state standards, particularly in the summer, and microbial water quality in the sloughs was generally lower than in ponds during both seasons. Specifically, the inflow of water from the sloughs to the ponds during the summer, more so than waterfowl use, appeared to increase the FIB concentrations in the ponds. The box model results suggested that the ponds served as net wetland sources and sinks for FIB, and high bird abundances in the winter likely contributed to net winter source terms for two of the three FIB in both ponds. Eight serovars of the human pathogen Salmonella were isolated from slough and pond waters, although the source of the pathogen to these wetlands was not identified. Thus, it appeared that factors other than bird abundance were most important in modulating FIB concentrations in these ponds. 相似文献

Microbial activity and distribution during enhanced contaminant dissolution from a NAPL source zone 总被引:1,自引:0,他引:1
Laboratory experiments were conducted to assess microbial reductive dechlorination in one-dimensional sand columns containing a 10 cm long source zone of uniformly distributed residual tetrachloroethene (PCE) nonaqueous phase liquid (NAPL), a 10 cm long transition zone directly down-gradient of the source zone containing some nonuniformly distributed NAPL ganglia, and a 40 cm long plume region down-gradient of the transition zone. The activity and distribution of Sulfurospirillum multivorans, a PCE-to-1,2-cis-dichloroethene (cis-DCE) dechlorinating bacterium, was evaluated in columns containing either a mixed-NAPL (0.25 mol/mol PCE in hexadecane) or pure PCE-NAPL. Significant dechlorination of PCE to cis-DCE was observed in the mixed-NAPL column, resulting in 53% PCE-NAPL mass recovery in the effluent with PCE-NAPL dissolution enhanced by up to 13.6-fold (maximum) and 4.6-fold (cumulative) relative to abiotic dissolution. Quantitative real-time PCR targeting pceA, the PCE reductive dehalogenase gene of S. multivorans, revealed that S. multivorans cells were present in the NAPL source zone, and increased in numbers (i.e., grew) throughout the source and transition zones. In contrast, minimal reductive dechlorination and microbial growth were observed in the column containing pure PCE-NAPL, where aqueous-phase PCE concentrations reached saturation. These results demonstrate that microbial growth within NAPL source zones is possible, provided that contaminant concentrations remain below levels toxic to the dechlorinating organisms, and that microbial growth can result in significant bioenhanced NAPL dissolution. 相似文献

The introduction of advanced welding methods as an alternative joining process to riveting in the manufacture of primary aircraft structure has the potential to realise reductions in both manufacturing costs and structural weight. However, welding processes can introduce undesirable residual stresses and distortions in the final fabricated components, as well as localised loss of mechanical properties at the weld joints. The aim of this research is to determine and characterise the key process effects of friction stir welding assembly methods on stiffened panel static strength performance. Utilising experimentally validated finite element modelling methods, it is demonstrated that welding induced residual stresses have a significant influence, and that collapse behaviour is less sensitive to advanced process effects and process effect magnitudes than panel initial buckling behaviour. 相似文献

An anaerobic mixed microbial culture was enriched from soil and groundwater taken from a site contaminated with trichloroethene (TCE). This enrichment culture was divided into four subcultures amended separately with either perchloroethene (PCE), TCE, cis-dichloroethene (cDCE) or vinyl chloride (VC). In each of the four subcultures, the chlorinated ethenes were rapidly, consistently, and completely converted to ethene at rates of 30-50 micromol/l of culture per day, or an average 160 micro-electron equivalents/l of culture per day. These cultures were capable of sustained and rapid dechlorination of VC, and could not dechlorinate 1,2-dichloroethane, differentiating them from Dehalococcoides ethenogenes, the only known isolate capable of complete dechlorination of PCE to ethene. Chloroform (CF) and 1,1,1-trichloroethane, frequent groundwater co-contaminants with TCE and PCE, inhibited chlorinated ethene dechlorination. Most strongly inhibited was the final conversion of VC to ethene, with complete inhibition occurring at an aqueous CF concentration of 2.5 microM. Differences in rates and community composition developed between the different subcultures, including the loss of the VC enrichment culture's ability to dechlorinate PCE. Denaturing gradient gel electrophoresis of amplified bacterial 16S rRNA gene fragments identified three different DNA sequences in the enrichment cultures, all phylogenetically related to D. ethenogenes. Based on the PCR-DGGE results and substrate utilization patterns, it is apparent that significant mechanistic differences exist between each step of dechlorination from TCE to ethene, especially for the last important dechlorination step from VC to ethene. 相似文献

In this study, the adsorption of bisphenol A (BPA) and 17α-ethinyl estradiol (EE2) from landfill leachate onto single-walled carbon nanotubes (SWCNTs) was investigated. Different leachate solutions were prepared by altering the pH, ionic strength, and dissolved organic carbon (DOC) in the solutions to mimic the varying water conditions that occur in leachate during the various stages of waste decomposition. The youngest and oldest leachate solutions contained varying DOC and background chemistry and were represented by leachate Type A (pH = 5.0; DOC = 2500 mg/L; conductivity = 12,500 μS/cm; [Ca2+] = 1200 mg/L; [Mg2+] = 470 mg/L) and Type E (pH = 7.5; DOC = 250 mg/L; conductivity = 3250 μS/cm; [Ca2+] = 60 mg/L; [Mg2+] = 180 mg/L). These solutions were subsequently combined in different ratios to produce intermediate solutions, labeled B-D, to replicate time-dependent changes in leachate composition. Overall, a larger fraction of EE2 was removed as compared to BPA, consistent with its higher log KOW value. The total removal of BPA and EE2 decreased in older leachate solutions, with the adsorptive capacity of SWCNTs decreasing in the order of leachate Type A > Type B > Type C > Type D > Type E. An increase in the pH from 3.5 to 11 decreased the adsorption of BPA by 22% in young leachate and by 10% in old leachate. The changes in pH did not affect the adsorption of EE2 in the young leachate, but did reduce adsorption by 32% in the old leachate. Adjusting the ionic strength using Na+ did not significantly impact adsorption, while increasing the concentration of Ca2+ resulted in a 12% increase in the adsorption of BPA and a 19% increase in the adsorption of EE2. DOC was revealed to be the most influential parameter in this study. In the presence of hydrophilic DOC, represented by glucose in this study, adsorption of the endocrine disrupting compounds (EDCs) onto the SWCNTs was not affected. In the absence of SWCNTs, hydrophobic DOC (i.e., humic acid) adsorbed 15-20% of BPA and EE2. However, when the humic acid and SWCNTs were both present, the overall adsorptive capacity of the SWCNTs was reduced. Hydrophobic (π-π electron donor-acceptor) interactions between the EDCs and the constituents in the leachate, as well as interactions between the SWCNTs and the EDCs, are proposed as potential adsorption mechanisms for BPA and EE2 onto SWCNTs. 相似文献

Zearalenone (ZON) is one of the worldwide most common mycotoxins and exhibits estrogenic activity in the range of natural steroid estrogens such as 17β-estradiol (E2). The occurrence of ZON has been reported in drainage water, soil, wastewater effluents and rivers, but its ecotoxicological effects on fish have hardly been investigated.In this study the estrogenic potency of the ZON was compared to E2 in a recombinant yeast estrogen screen (rYES) and the effects of waterborne ZON exposure on reproduction, physiology and morphology of zebrafish (Danio rerio) were investigated in a 42-day reproduction experiment. E2 as well as ZON evoked a sigmoid concentration-response curve in the rYES with a mean EC50 of 2 and 500 μg/L, respectively, resulting in an E2:ZON EC50 ratio of 1:250. Exposure to ZON for 21 days reduced relative spawning frequency at 1000 and 3200 ng/L to 38.9 and 37.6%, respectively, and relative fecundity at 100, 320, 1000 and 3200 ng/L to 74.2, 41.7, 43.8 and 16.7%, respectively, in relation to the 21-day pre-exposure period. A 4.4 and 8.1 fold induction of plasma vitellogenin (VTG) was observed in male zebrafish at 1000 and 3200 ng/L ZON, respectively. Exposure to ZON did not affect fertility, hatch, embryo survival and gonad morphology of zebrafish.The results of this study demonstrate that although ZON possesses a moderate estrogenic potency in vitro, it exhibits a comparably strong effect on induction of VTG and reproduction in vivo. This indicates that ZON might contribute to the overall estrogenic activity in the environment and could therefore pose a risk for wild fish in their natural habitat. 相似文献

Recently developed methods to calculate the time required for ceiling mounted heat and smoke detectors to respond to growing fires are reviewed. A computer program that calculates activation times for both fixed temperature and rate of rise heat detectors in response to fires that increase in heat release rate proportionally with the square of time from ignition is given. This program produces nearly equivalent results to the tables published in Appendix C, Guide for Automatic Fire Detector Spacing (NFPA 72E, 1984). A separate method and corresponding program are provided to calculate response time for fires having arbitrary heat release rate histories. This method is based on quasi-steady ceiling layer gas flow assumptions. Assuming a constant proportionality between smoke and heat released from burning materials, a method is described to calculate smoke detector response time, modeling the smoke detector as a low temperature heat detector in either of the two response time models.Nomenclature A

Viral hepatitis type A is the most prevalent waterborne disease in U.S.A. and the number of cases has been increasing in recent years (Melnick et al., 1978). Till now, few works were performed on Hepatitis A Virus (HAV) concentration in water (Hejkal et al., 1982, Elkana et al., 1983). In this paper the filter adsorption-elution method, widely used with other viruses (Wallis and Melnick, 1967; Fields et Metcalf, 1975; Smith and Gerba, 1982), was selected to concentrate HAV from experimentally-contaminated distilled water. Filtration was performed through two cellulose membranes (RAWP 04700 and HAWP 04700 Millipore). Eluate or filtrate viruses were precipitated by polyethylene glycol 6000. HAV is detected by solid phase radioimmunoassay according to a previously described method (Deloince et al., 1982). Figure 1 shows the results of adsorbing HAV at different pH levels. Analysis of variance pointed out that, for the pH levels between 3 and 4, the differences were not significant (P = 0.05). Table 1 shows the effects of pH and number of passages of 3% beef extract or 0.2 M glycin buffer on HAV elution. For both eluents the differences in function of pH levels and number of passages of eluent through the membrane were not significant (variance analysis P = 0.05). On the other hand, the comparison between the two eluents pointed out the higher efficiency of beef extract (t-test, P = 0.001). After this procedure, HAV replication occurred in human hepatocarcinoma cells PLC/PRF/5 (Fig. 2) inoculated according to a previously described method (Crance et al., 1983). As poliovirus (Sobsey et al., 1973), HAV adsorption is the most efficient at acid pH levels. Between pH 4.5 and 5, the remarkable adsorption decrease might be related to the inversion of the HAV electrostatic charges sign, i.e. to the occurrence of an isoelectric point. The HAV elution is different from the poliovirus one, the efficiency of which increased with pH (Wallis et al., 1972) and eluent passage numbers (Farrah et al., 1976). That might suggest that from pH 7.5, both the HAV particles and membranes have negative charges which give enough repulsive forces to allow a good elution efficiency. Improvement of HAV elution efficiency by beef extract is similar to a result obtained with poliovirus by Landry et al. (1978). This effect might be explained by the proteinous material presence in beef extract. HAV replication in cell cultures shows the biological properties preservation after this procedure. 相似文献

Geochemical mass balances were computed for water years 1992-1997 (October 1991 through September 1997) for the five watersheds of the U.S. Geological Survey Water, Energy, and Biogeochemical Budgets (WEBB) Program to determine the primary regional controls on yields of the major dissolved inorganic solutes. The sites, which vary markedly with respect to climate, geology, physiography, and ecology, are: Allequash Creek, Wisconsin (low-relief, humid continental forest); Andrews Creek, Colorado (cold alpine, taiga/tundra, and subalpine boreal forest); Río Icacos, Puerto Rico (lower montane, wet tropical forest); Panola Mountain, Georgia (humid subtropical piedmont forest); and Sleepers River, Vermont (humid northern hardwood forest). Streamwater output fluxes were determined by constructing empirical multivariate concentration models including discharge and seasonal components. Input fluxes were computed from weekly wet-only or bulk precipitation sampling. Despite uncertainties in input fluxes arising from poorly defined elevation gradients, lack of dry-deposition and occult-deposition measurements, and uncertain sea-salt contributions, the following was concluded: (1) for solutes derived primarily from rock weathering (Ca, Mg, Na, K, and H(4)SiO(4)), net fluxes (outputs in streamflow minus inputs in deposition) varied by two orders of magnitude, which is attributed to a large gradient in rock weathering rates controlled by climate and geologic parent material; (2) the net flux of atmospherically derived solutes (NH(4), NO(3), SO(4), and Cl) was similar among sites, with SO(4) being the most variable and NH(4) and NO(3) generally retained (except for NO(3) at Andrews); and (3) relations among monthly solute fluxes and differences among solute concentration model parameters yielded additional insights into comparative biogeochemical processes at the sites. 相似文献

Library-dependent (LD) (biochemical fingerprinting of Escherichia coli and enterococci) and library-independent (LI) (PCR detection of human-specific biomarkers) methods were used to detect human faecal pollution in three non-sewered catchments. In all, 550 E. coli isolates and 700 enterococci isolates were biochemically fingerprinted from 18 water samples and compared with metabolic fingerprint libraries of 4508 E. coli and 4833 enterococci isolates. E. coli fingerprints identified human unique biochemical phenotypes (BPTs) in nine out of 18 water samples; similarly, enterococci fingerprints identified human faecal pollution in 10 water samples. Seven samples were tested by PCR for the detection of biomarkers. Human-specific HF134 Bacteroides and enterococci surface protein (esp) biomarkers were detected in five samples. Four samples were also positive for HF183 Bacteroides biomarker. The combination of biomarkers detected human faecal pollution in six out of seven water samples. Of the seven samples analysed for both the indicators/markers, at least one indicator/marker was detected in every sample. Four of the seven PCR-positive samples were also positive for one of the human-specific E. coli or enterococci BPTs. The results indicated human faecal pollution in the studied sub-catchments after storm events. LD and LI methods used in this study complimented each other and provided additional information regarding the polluting sources when one method failed to detect human faecal pollution. Therefore, it is recommended that a combination of methods should be used to identify the source(s) of faecal pollution where possible. 相似文献
